I don't know if there is a list of the actors/actresses who have had bit parts in the show and have gone on to become relatively big hits
heres two obvious ones:
Courtney Cox - played Jerry's "wife", she wanted a dry cleaning discount. Went on to "Friends"
Debra Messing - played Jerry's girlfirend in the yadda yadda episode (who needs the blacks and the Jews). Now starring in "Will and Grace"

I had already seen her on Lois & Clark when I saw her on Seinfeld. Seinfeld was apparently older than I thought when I first started watching it, so I must have seen a re-run of Seinfeld when I saw her for the first time.
I loved Lois & Clark for the first season or so. I recall seeing the first show and thinking "This has got to be a hit". I don't think I have ever thought that about any other show and been right in my prognosis.
