In this weeks Warner Bros. Friends newsletter, it is said that Rachel only has one sister:

Yes, this is what I thought--middle sister. Btw, the eldest sister has not appeared on the show. So perhaps the "one sister" thing was about her younger sister, Jill who dated Ross briefly.

In one of the episodes in the Best of Friends vol 1 or 2 (sorry haven't watched them in a while, not sure which one), they show a scene not originally aired on tv where her sister asks her if she slept with this guy because he yelled out 2 for 2 during a passionate moment...it wasn't Jill. So...unless they made a major error that would pretty much confirm she had 2 sisters.
I am new here so I am not sure if it has already been said, but there was one episode where someone asked something about her sisters and she said "yeah, but they aren't nice people" so I am assuming that she has at least 2 sisters.

In the fifth season, TOW The Inappropriate Sister, Rachel talks with Danny about sibling relationships and she says, "I have two sisters, but one of them is a little manish."
